Abstract

This study investigated the transovarial detection of dengue virus in Aedes aegypti pupae in Dasmarinas City, Cavite. Ovitraps were placed along Aguinaldo highway and collected for pooling pupal samples after 7 days. The mosquitoes were distinguished through morphological characteristics under microscopy. Relative humidity was taken for each sample site using sling psychrometer. Identified Aedes aegypti pupa were macerated and put in an Eppendorf tube for Real Time – Reverse Transcriptase Polymerase Chain Reaction. The results show that there is a significant negative correlation between the month and relative humidity (P=0.0000) that resulted in the increase in the average number of larva. A positive correlation (P=0.0326) between the sites and the number of larvae is also observed showing the site with the highest number of larva collected per month i.e. Site 5 (Kin Yang School, Sabang) for July, Site 4 (Augustin Grove Village) for August and Site 10 (Villa Isabel) for September. No transovarial transmission is detected in all of the samples using real time – RT PCR. The study concludes that the development of larva and pupa are dependent on relative humidity and transovarial transmission is not possible. This study implied that transovarial transmission will not occur in the wild therefore dengue epidemiological surveillance should focus on the adult Aedes mosquito.
