Abstract

The cytotoxic effect of crude leaf extracts of Jade vine (S. macrobotrys) and Lagundi (Vitex negundo) has been evaluated against Vero cells and HeLa cells. Different concentrations of crude leaf extracts were applied to cultured cells and incubated for 72 hours. Cell survival was evaluated using MTT assay, a rapid and sensitive spectrophotometric assay for determining viability in monolayer cultured cell lines, with specific applications in normal and cancer cell lines determinations. The crude leaf extracts of Jade vine (S. macrobotrys) and Lagundi (Vitex negundo) showed a toxic effect against cultured cell lines. The effective concentration of the HeLa cells against Jade vine (S. macrobotrys) and Lagundi (Vitex negundo) extract is higher than the effective concentration of Vero cells. In conclusion, for a plant extract to be considered anti-cancer, the EC50 of the Hela cells must be lower than the EC50 of the Vero cells. In this case, the Hela cells proved to have a much higher EC50 as opposed to the Vero cells which have an EC50 establishing the fact that Jade vine (S. macrobotrys) and Lagundi (Vitex negundo) are not anti-cancer.
